Comparison of Hb A10 Measurment by VariantTM, Ion-Exchange High Performance Liquid Chromatography and IMxR, Ion Capture Assay




Abstract
Background:
Glycosylated hemoglobin(Hb A1c) is a useful indicator for measuring the long term glucose level in diabetes mellitus patients. Thus Hb A1c has been used to monitor the patients' compliance of antidiabetetic therapy. Various methods are
available to
measure Hb A1c, Among them IMxR(Abbott Laboratories, IL, USA), an ion capture assay device, has widely been used in assaying Hb A1c since this equipment is popular for number of immunochemical assays. However, IMxR is not suitable for the large
volume
laboratories. Therefore, performance of a fast throughput instrument for measuring Hb A1c, VariantTM(Bio-Rad Inc, CA, USA), an ion-exchange high performance liquid chroatography, has been evaluated and compared wit that of IMxR.
Methods:
One hundred thirty four randomly selected fresh whole blood specimens have been subjected to hb A1c assays uring VariantTM and IMxR. The results from two equipments were compared statistically by using paried t-test and linear regression
model.
Results:
The mean¡¾standard deviation of Hb A1c for VariantTM was 8.00¡¾2.39% and for IMxR 7.72¡¾2.09% ; and these were significantly different(p=0.0024). The regression equation was %Hb A1c VariantTM=1.024. IMxR+0.086(r=0.804). The cofficient of
variation
were 6.0%(Hb A1c level 5.8%) and 4.3%(hb A10 level 12.6%) for IMxR.
Conclusions:
VariantTM was precise but it showed on showed on an average 3.6% higher values than that obtained by IMxR. However, the VariantTM may save labor cost considerably.
